Background: RNF111 (Ring finger protein 111) acts in the NODAL pathway of mesoderm patterning during embryonic development. It is an E3 ubiquitin-protein ligase which promotes the ubiquitination of inhibitory SMADs such as SMAD7, induces their proteasomal degradation and thereby enhances the transcriptional activity of TGF-beta and BMP. It also induces degradation of the transriptional repressors SnoN and c-Ski. RNF111, therefore, effectively enhances TGF-beta signaling through simultaneous down-regulation of two distinct types of negative regulators, Smad7 and SnoN/c-Ski.
